1. Gamasina mite communities (Acari: Mesostigmata) in grain production systems of the southwestern Brazilian Amazon. [2021]

Abstract
Emphasis has been given in the Brazilian state of Mato Grosso to the implementation of more sustainable production practices, including what has been termed agrosilvopastoral production system (ASPS), which involves the integration of different types of land use in a same area along time, seeking increased ecological stability. The Gamasina mite group (order Mesostigmata) is usually diverse and abundant in non-cultivated soils, where they can prey on arthropods and nematodes; some are commercialized for biological pest control. The objective of this study was to evaluate the diversity and abundance of edaphic Gamasina in plots of grain production managed under conventional (alternating cultivation) system and under ASPS, both with the adoption of no-tillage cultivation, in the municipality of Sinop, Mato Grosso state. In samples of soil and litter taken biweekly from August 2015 to May 2016, 762 Gamasina representing 32 species of 21 genera and nine families were collected. Considering both systems together, the dominant Gamasina were quite different from those of other parts of Brazil. Calculated ecological indexes showed no major differences between the two systems, possibly because of the relatively short time since the implementation of ASPS. In both systems, Rhodacaridae was one of the dominant families (37.5% of the Gamasina), followed by Macrochelidae (20.9%) and Laelapidae (18.8%). The most abundant species were Multidentirhodacarus squamosus Karg (Rhodacaridae), a new species of Holostaspella (Macrochelidae) and Cosmolaelaps barbatus Moreira, Klompen and Moraes (Laelapidae). Future studies are warranted, allowing more time for the adoption of ASPS to produce possible ecological changes. [ABSTRACT FROM AUTHOR]

Abstract
A lagarta de Spodoptera frugiperda (J. E. Smith) é uma das principais pragas do milho e para seu controle o Bacillus thuringiensis se destaca por sua atividade entomopatogênica. Este trabalho objetivou a caracterização molecular de isolados de B. thuringiensis quanto à presença do gene cry1 e a avaliação da sua eficiência no controle de lagartas de S. frugiperda. Nas análises da PCR, foi utilizado o Gral-cry1 para confirmação da presença do gene cry1 nos 115 isolados. Uma suspensão de 3 x 10(8) esporos/ml banhou a dieta utilizada para alimentação de 30 lagartas por isolado, com três repetições. A identificação do tipo de genes cry1 dos diferentes isolados foi realizada para cinco sub-classes de genes e análises de regressão linear foram realizadas para verificar possíveis associações entre a presença de um gene cry individual e altos níveis de toxicidade. Ttodos os DNAs amplificados com os iniciadores Gral-cry1 apresentaram produto de amplificação com tamanho esperado. Quanto aos níveis de eficiência inseticida contra a lagarta-do-cartucho, 41 isolados apresentaram 100% de mortalidade e 16 apresentaram índice entre 75% e 90%. O gene cry1Ab esteve presente em 80 isolados, cryB em 69 isolados, cry1Ac em todos os isolados, cryV e cry1E em 93 e 27 isolados, respectivamente. Os valores referentes ao efeito individual de cada gene na mortalidade de larvas foram significativos a 1% de probabilidade, para os genes cry1Ac e cry1E.
The fall armyworm, Spodoptera frugiperda (J. E. Smith), is one of the main corn pests and Bacillus thuringiensis is important in its control because of its entomopathogenic property. The objective of this study was the molecular characterization of B. thuringiensis isolates for cry1 locus presence and the assessment of the efficiency of these isolates in controlling S. frugiperda caterpillars. Gral-cry1 was used in the PCR analyses to confirm the presence of the cry1 locus in 15 isolates. A 3 x 10(8) spore/ml suspension bathed the diet used to feed 30 caterpillars per isolate, with three replications. The cry1 locus type genes of the different isolates were identified for five gene subclasses; linear regression analyses were carried out to ascertain possible associations between the presence of an individual cry1 locus gene and high levels of toxicity. All the DNAs amplified with Gral-cry1 presented an amplification product with the expected size. Regarding the levels of insecticide efficiency against the cob worm, 41 isolates presented 100% mortality and 16 presented an index between 70% and 90%. The cry1Ab gene was present in 80 isolates, cryb in 69 isolates, cry1Ac in all the isolates and cryv and cry1E in 93 and 27 isolates, respectively. The values regarding the individual effect of each gene on caterpillar mortality were significant at 1% probability for the cry1Ac and cry1E genes.

Abstract
BACKGROUND: Supernatant-secreted proteins in Bacillus thuringiensis (Berliner) (Bt) with insecticidal activity provide an important source of information for discovery of new useful strains and/or entomotoxins. However, physiological variation among isolates might interfere in the detection efficiency of screening procedures on Bt collections. The aim of this study was to assess the magnitude of this variation in a sample of isolates from a tropical Bt collection, which was gauged through the assessment of their temporal patterns of growth and protein secretion in culture supernatants (SNs), as well as of the corresponding toxicity against fall armyworm (Spodoptera frugiperda,JE Smith). Feeding bioassays were performed, with larvae being treated with heated and non-heated total protein extracted from SNs collected at different culture times. Larva mortality and reduction in pupa formation were observed. RESULTS: Intra- and interisolate variations were observed in the temporal patterns of growth, quality and quantity of protein secreted, as well as in insecticidal activity of these SNs, based on larvae mortality and pupation rates. These results suggest that the insecticidal potential of certain isolates can be hidden if comparisons are done on the basis of the same number of cells in the culture and/or the same culturing time. CONCLUSIONS: Methods of screening Bt collections on the basis of feeding bioassays can be misleading with regards to identifying more promising isolates for biocontrol purposes if physiological differences are not considered. The consequences and implications of these findings for the development of experimental systems that depend on toxicity bioassays to identify alternative Bt strains and entomotoxins with practical applicability have been discussed.

Abstract
The total of 22 Baculovirus isolates surveyed in different corn producing regions in Brazil were used against fall armyworm, Spodoptera frugiperda (J.E. Smith). The viruses were purified and their suspensions were used to feed fall armyworm larvae from 4th and 5th instar. The mortality rate was checked daily and the infected larvae were frost after death, what generally occurred between the 5th and 7th day after virus ingestion. The 22 Baculovirus isolates were used in six concentrations (from 10³ to 10(8) polyhedra/ml) and one check treatment with water. Mortality rate, larval period, pupal period, pupa weight and lethal concentration (LC50) were determined for all isolates. Significant differences were found among all isolates and different concentrations, also interaction between isolate x virus concentration for all characteristics evaluated, except for pupal period. Amplification patterns of 54 RAPD markers, being 41 polymorphic among the isolates, were used to evaluate the genetic distance and its correlation with the fall armyworm larvae mortality rate. The genetic diversity calculated by the Jaccard's coefficient using the molecular data allowed a division of the isolates into two groups, with a high level of confidence. These groups did not present any association with the mortality rate caused by the isolates or with their geographical distribution. However, a RAPD fragment OPW04.2280 was highly associated with the larvae mortality rate and with LC50, explaining 23 and 65% of the phenotypic variation for these traits among the isolates, respectively.
Foram utilizados 22 isolados de vírus amostrados em diferentes regiões produtoras de milho do Brasil. Os vírus foram purificados e suas suspensões fornecidas a lagartas sadias do 3º e 4º ínstar de Spodoptera frugiperda (J.E. Smith). A mortalidade foi avaliada diariamente, e as lagartas infectadas foram congeladas logo após sua morte, o que em geral ocorreu do 5º ao 7º dia após ingestão do vírus. Os isolados foram usados em seis concentrações (10³ a 10(8) poliedros/ml) e uma testemunha (água). Os percentuais de mortalidade, duração do período larval e período pupal, peso de pupa e a concentração letal (CL50) foram determinados para todos os isolados. Foram observadas diferenças significativas entre todos os isolados e concentrações testadas para todos os parâmetros avaliados, e também foi constatada a presença da interação isolado x concentração, exceto para período pupal. Os padrões de amplificação de 54 marcadores RAPD, sendo 41 polimórficos, foram utilizados para avaliar a distância genética e a sua correlação com os índices de mortalidade das lagartas. A divergência genética calculada pelo coeficiente Jaccard utilizando os dados moleculares permitiu dividir os isolados em dois grupos, com um elevada confiabilidade. O agrupamento não apresentou associação com a taxa de mortalidade causada pelos isolados ou com sua distribuição geográfica. No entanto, um fragmento de RAPD OPW04.2280 apresentou-se altamente associado com a mortalidade das lagartas e com a CL50, explicando 23% e 65% da variação fenotípica para essas características entre os isolados virais, respectivamente.

Abstract
Novel vegetative insecticidal proteins (Vips) identified in the supernatant of Bacillus thuringiensis (B.t.) cultures have shown to provide adequate control over a wide spectrum of economically important crop pests. To evaluate the potential applicability of these proteins against fall armyworm (Spodoptera frugiperda Smith) larvae, the most important insect pest for tropical maize, the characteristics and mortality effects of culture supernatants from five B.t. strains were investigated. Striking differences among strains were detected, not only in terms of efficiency in killing the insect, but also regarding to mortality effects of heated and non-heated supernatants, which were used to distinguish the heat-sensitive protein-derived insecticidal fraction from a thermostable one, with a non-protein nature (b-exotoxinas). The qualitative, quantitative and temporal patterns of total protein secretion in the medium (supernatant) were assessed through spectrophotometry and polyacrylamide gel electrophoresis. The strains showed remarkably distinct rates of growth and timing for protein secretion relative to cell density in culture. Moreover, the electrophoretic-banding patterns also varied in a strain-specific manner, both in denaturing and non denaturing conditions. Polypeptides displaying a molecular weight that is very close to the expected for previously identified Vip3A proteins were found for the strains with high supernatant-mortality ratios. The data suggest the feasibility and usefulness of searching for protein-derived (Vip-like) insecticidal fractions in B.t. supernatants as a mean of developing especific and efficient alternatives of biological control to be employed in integrated pest management programs of S. frugiperda in tropical maize.
Recentemente, proteínas inseticidas vegetativas (Vips), identificadas no sobrenadante da cultura de certas cepas de Bacillus thuringiensis (B.t.), foram capazes de controlar alguns insetos-praga economicamente importantes. Como avaliação do potencial de aplicabilidade destas proteínas contra a lagarta-do-cartucho (Spodoptera frugiperda Smith), o inseto-praga mais importante da cultura do milho, as características e o efeito de mortalidade do sobrenadante da cultura de cinco cepas de B.t. foram investigadas. Notáveis diferenças entre as cepas foram detectadas, não somente em termos de eficiência na mortalidade dos insetos, mas também nas diferenças de mortalidade entre os sobrenadantes aquecido e não-aquecido, tratamentos estes que foram usados para distinguir a fração protéica inseticida termo-sensível de outra fração termo-estável, de natureza não-protéica (b-exotoxinas). Os padrões qualitativo, quantitativo e temporal de secreção da proteína total no meio (sobrenadante) foram avaliados por espectrofotometria e eletroforese em gel de poliacrilamida. As cepas apresentaram distintas taxas de crescimento e tempos de secreção de proteínas em relação a densidade de células na cultura. Além disso, o padrão de bandas eletroforéticas também variou de maneira cepa-específica, em ambas condições desnaturantes e não-desnaturantes. Polipeptídeos que apresentaram peso molecular semelhantes aos esperados para proteínas Vip3A identificadas anteriormente foram encontrados em cepas com alta taxa de mortalidade do sobrenadante. Os dados sugerem a possibilidade e a utilidade de pesquisar frações inseticidas de natureza protéica (do tipo Vip) em sobrenadantes de B.t., como forma para desenvolver alternativas específicas e eficientes de controle biológico que possam ser empregadas em programas de manejo integrado de S. frugiperda em milho tropical.

Abstract
A total of 3408 strains of Bacillus thuringiensis were collected from 1448 soil samples in 10 Brazilian states, four different geographical regions, covering 96 counties. These strains were evaluated against Spodoptera frugiperda (J.E. Smith) larvae. Only 62% killed between 81% and 100% and 1758 caused no mortality. Highest proportion of efficient strains (larval mortality above 75%) was found from the total isolated per region in the South Region (16.6%), followed by Western Central (3.1%), Southeast (1.1%) and Northeast Region (0.4%).
O total de 3408 cepas de Bacillus thuringiensis foi isolado de 1448 amostras de solo provenientes de 10 estados, em quatro regiões brasileiras, abrangendo 96 municípios. As cepas foram avaliadas contra lagartas de Spodoptera frugiperda (J. E. Smith) sendo que apenas 62% mataram entre 81% e 100%, e 1758 não causaram mortalidade. O Sul destacou-se com 16,6% de cepas eficientes (mortalidade>75%) do total obtido por região, seguido das regiões Centro Oeste (3,1%), Sudeste (1,1%), e Nordeste (0,4%).

Abstract
Little is known about the jumping plant-lice of Brazil from where seven families, 45 genera and 76 species have been previously reported, but estimates suggest that there may be as many as 1,000 species. This study reports 34 species of Psylloidea which were collected along the edges of Amazon-Cerrado natural transitional forests in the municipality of Sorriso, state of Mato Grosso, from August 2013 to July 2014. E. Smith) é uma das principais pragas do milho e para seu controle o Bacillus thuringiensis se destaca por sua atividade entomopatogênica. Este trabalho objetivou a caracterização molecular de isolados de B. thuringiensis quanto à presença do gene cry1 e a avaliação da sua eficiência no controle de lagartas de S. frugiperda. Nas análises da PCR, foi utilizado o Gral-cry1 para confirmação da presença do gene cry1 nos 115 isolados. Uma suspensão de 3 x 10(8) esporos/ml banhou a dieta utilizada para alimentação de 30 lagartas por isolado, com três repetições. A identificação do tipo de genes cry1 dos diferentes isolados foi realizada para cinco sub-classes de genes e análises de regressão linear foram realizadas para verificar possíveis associações entre a presença de um gene cry individual e altos níveis de toxicidade. Ttodos os DNAs amplificados com os iniciadores Gral-cry1 apresentaram produto de amplificação com tamanho esperado. Quanto aos níveis de eficiência inseticida contra a lagarta-do-cartucho, 41 isolados apresentaram 100% de mortalidade e 16 apresentaram índice entre 75% e 90%. O gene cry1Ab esteve presente em 80 isolados, cryB em 69 isolados, cry1Ac em todos os isolados, cryV e cry1E em 93 e 27 isolados, respectivamente. Os valores referentes ao efeito individual de cada gene na mortalidade de larvas foram significativos a 1% de probabilidade, para os genes cry1Ac e cry1E. The fall armyworm, Spodoptera frugiperda (J. E. Smith), is one of the main corn pests and Bacillus thuringiensis is important in its control because of its entomopathogenic property. The objective of this study was the molecular characterization of B. thuringiensis isolates for cry1 locus presence and the assessment of the efficiency of these isolates in controlling S. frugiperda caterpillars. Gral-cry1 was used in the PCR analyses to confirm the presence of the cry1 locus in 15 isolates. A 3 x 10(8) spore/ml suspension bathed the diet used to feed 30 caterpillars per isolate, with three replications. The cry1 locus type genes of the different isolates were identified for five gene subclasses; linear regression analyses were carried out to ascertain possible associations between the presence of an individual cry1 locus gene and high levels of toxicity. All the DNAs amplified with Gral-cry1 presented an amplification product with the expected size. Regarding the levels of insecticide efficiency against the cob worm, 41 isolates presented 100% mortality and 16 presented an index between 70% and 90%. The cry1Ab gene was present in 80 isolates, cryb in 69 isolates, cry1Ac in all the isolates and cryv and cry1E in 93 and 27 isolates, respectively. The values regarding the individual effect of each gene on caterpillar mortality were significant at 1% probability for the cry1Ac and cry1E genes. Abstract
The total of 22 Baculovirus isolates surveyed in different corn producing regions in Brazil were used against fall armyworm, Spodoptera frugiperda (J.E. Smith). The viruses were purified and their suspensions were used to feed fall armyworm larvae from 4th and 5th instar. The mortality rate was checked daily and the infected larvae were frost after death, what generally occurred between the 5th and 7th day after virus ingestion. The 22 Baculovirus isolates were used in six concentrations (from 103 to 108 polyhedra/ml) and one check treatment with water. Mortality rate, larval period, pupal period, pupa weight and lethal concentration (LC50) were determined for all isolates. Significant differences were found among all isolates and different concentrations, also interaction between isolate x virus concentration for all characteristics evaluated, except for pupal period. Amplification patterns of 54 RAPD markers, being 41 polymorphic among the isolates, were used to evaluate the genetic distance and its correlation with the fall armyworm larvae mortality rate. The genetic diversity calculated by the Jaccard's coefficient using the molecular data allowed a division of the isolates into two groups, with a high level of confidence. These groups did not present any association with the mortality rate caused by the isolates or with their geographical distribution. However, a RAPD fragment OPW04.2280 was highly associated with the larvae mortality rate and with LC50, explaining 23 and 65% of the phenotypic variation for these traits among the isolates, respectively.. 2005

Abstract
Recentemente, proteinas inseticidas vegetativas (Vips), identificadas no sobrenadante da cultura de certas cepas de Bacillus thuringiensis (B.t.), foram capazes de controlar alguns insetos-praga economicamente importantes. Como avaliacao do potencial de aplicabilidade destas proteinas contra a lagarta-do-cartucho (Spodoptera frugiperda Smith), o inseto-praga mais importante da cultura do milho, as caracteristicas e o efeito de mortalidade do sobrenadante da cultura de cinco cepas de B.t. foram investigadas. Notaveis diferencas entre as cepas foram detectadas, nao somente em termos de eficiencia na mortalidade dos insetos, mas tambem nas diferencas de mortalidade entre os sobrenadantes aquecido e nao-aquecido tratamentos estes que foram usados para distinguir a fracao proteica inseticida termo-sensivel de outra fracao termo-estavel, de natureza nao-proteica (b-exotoxinas). Os padroes qualitativo , quantitativo e temporal de secrecao da proteina total no meio (sobrenadante) foram avaliados por espectrofotometria e eletroforese em gel de poliacrilamida. As cepas apresentaram distintas taxas de crescimento e tempos de secrecao de proteinas em relacao a densidade de celulas na cultura. Alem disso, o padrao de bandas eletroforeticas tambem variou de maneira cepa-especifica, em ambas condicoes desnaturantes e nao-desnaturantes. Polipeptideos que apresentaram peso molecular semelhantes aos esperados para proteinas Vip3A identificadas anteriormente foram encontradas em cepas com alta taxa de mortalidade do sobrenadante. Os dados sugerem a possibilidade e a utilidade de pesquisar fracoes inseticidas de natureza proteica (do tipo Vip) em sobrenadantes de B.t., como forma para desenvolver alternativas especificas e eficientes de controle biologico que possam ser empregadas em programas de manejo integrado de S. frugiperda em milho tropical. 1999

Abstract
Spermologus rufus Boheman (Coleoptera: Curculionidae) is an insect which attacks cacao nuts, oleaginous pits and Araucaria angustifolia (Bert.) Kuntze seeds. In this study the reproductive behavior of S. rufus was investigated to provide useful knowledge to support control measures of this insect. Research was carried out under laboratory conditions. Twenty-two couples of newly emerged adults were individualized in Wheaton flasks containing A. angustifolia seeds for oviposition and feeding. Mating was observed every day until the insects died. Females oviposited in the seeds; 80.4% of these seeds had only one egg. Eggs were mostly found in the median and more enlarged region of the seed and at the insertion point. Pre-oviposition period was, on average, 11.1±7.5 days and the oviposition period lasted 118.1±42.3 days. Most of the egg laying occurred between 0:00 and 8:00 am. Eggs were laid isolated and each female laid, on average, 146.7±43.9 eggs. After 60, 120 and 240 days after emergence, respectively, 80%, 90% and 10% of virgin females copulated and generated descendants, in relation to the control, where 90% copulated and generated descendants.

Abstract
Larvae and adults of Spermologus rufus Boheman can cause severe damage to seeds of Brazilian pine, Araucaria angustifolia. In this study, yeasts were isolated from seeds of A. angustifolia and from the digestive tract of S. rufus to verify their similarity and determine the existence of feeding sites in order to elaborate an artificial diet for this insect. Pichia spp. were found in seeds and Pichia spp. and P. stipitis Pignal in the digestive tract of S. rufus. The low variability among observed microorganisms suggests a food preference, which can facilitate the elaboration of artificial diets.
